"I am a short, not in DNDN, but a short seller nonetheless. I normally would not post in reference to a long, but this was wild stuff from an old short-seller's perspective. I have friends who are long DNDN, so I have been watching the action over the past couple of weeks. Ive been in this biz a looooong time, and I can honestly say that I have never seen anything in a listed stock like what happened today. That being said, albeit extremely rare, today's action was legit. I find it somewhat ironic that I again have been reading "The Black Swan" as this highly improbable scenario played out.

If you check the volume going into the lunch hour today in 3 minute intervals (approximately the duration of the selloff), then you see that the 3-minute volume had dropped from maybe 1.5 million shares during the busy morning to 150k shares going into lunch, to 60-90k shares in the minutes leading up to the selloff. All of my friends had stop losses in place. From reading these boards, so did a WHOLE LOT of other people. Expand this thought process across the entire market. There were millions of shares worth of stop loss orders sitting at various prices. Most traders will use a Day Low price or a Tick below Yesterday's Close to trigger their stop loss. Usually an effective strategy. However, doomsday material when everyone on Earth has the same idea. The last 10k+ block traded at $21.49 (close to DayLow). After that, the bids went off a cliff. The next +10k block didnt occur until sub $10. As the price dropped, the stop loss orders that were triggered become active market sell orders. Now, go back to the reference to the volume leading up to this. 30-60k per 3 minute interval. For whatever reason (and my gut tells me that someone planned this - its legal, and if they did, then they are among the trading legends with this move), sellers entered the market just enough to begin driving down the price a little bit. And a little is all it took. Over the next 3 minutes it can be guesstimated that several hundred thousand shares worth of stop losses were triggered - massive sell volume relative to the bids that were in place. I imagine a lot of this was attributable to the fact that everyone was on the sidelines at 1:26pm, awaiting the presentation at 1:30. Large selling with no buying became reminiscent of a bad penny stock when the manipulator leaves the bid. I am going through the floor feed right now and looking at the BestBid and BidSize for each second. There were no sizeable bids. The sell orders just chewed them apart. Lower prices triggered more market sell orders, and so the snowball grew.

Truly amazing. Any sane trader would have had that protection in place. And it was that very protection that screwed everyone. I would love to know if a group of shorts saw this opportunity. It was written on the wall for those smart enough to see it. Hats off if someone out there really pulled that off ... or was it simply a Black Swan event? The world may never know, but it is surely a damn good lesson on how even the best laid plans can go awry.

Sorry for anyone who lost on those stop losses. You played it right, but my gut tells me someone out there played you.

If it is any consolation, the shorts who did not cover in that debacle are sweating tonight. The margin man will come calling for many of them tomorrow."

The disease, which broke out in Mexico just days ago, has spread to Europe and testing of suspected cases was underway around the world. Governments everywhere have toughened their precautions and the World Health Organization raised its alert level from three to four, which is just two steps short of it declaring a full pandemic.

Though all 150 suspected deaths and most of the 2,000 or infections have been seen in Mexico, investors around the world have decided to run for cover, abandoning riskier assets such as stocks and diving back into safe haven assets like the dollar and the yen.

"Equity markets have continued to sell off amid worries that the spread of the virus will undermine hopes of an economic recovery later this year," said Stuart Bennett, an analyst at Calyon Credit Agricole.

"Until further details are available with regard to the spread of the virus and its threat, concerns will remain at the forefront signaling that risk aversion will remain elevated," he added.

With all the uncertainty gripping the markets, Wall Street futures were pointing to a sharply lower opening. Dow futures were down 103 points, or 1.3 percent, at 7,899 while the broader Standard & Poor's 500 futures fell 13.70 points, or 1.6 percent, to 843.10.

Earlier, Asia's markets took a pummeling with Japan's Nikkei index closing down 232.57 points, or 2.7 percent, to 8,493.77 and Hong Kong's Hang Seng ended 285.31 points, or 1.9 percent, lower at 14,555.11.

For the second day running, airlines and travel-related companies felt the brunt of the selling pressure. In Europe, Air France-KLM and Deutsche Lufthansa AG fell another 3 percent while British Airways PLC slumped a further 6 percent.

And in a repeat of Monday, pharmaceutical stocks, particularly those with high-profile anti-flu vaccines -- Switzerland's Roche Holding AG and GlaxoSmithkline PLC -- benefited amid the pandemic fears.

A potential pandemic wasn't the only distraction for investors, already uneasy about the results of the U.S. government's stress tests to gauge the health of the largest 19 banks.

The reports are set for release Monday, though Bank of America Corp. and Citigroup Inc. have been told by regulators the two will likely need to raise more capital, according to a Wall Street Journal report. The report suggested that Bank of America's capital shortfall could run into billions of dollars, which, in the current environment would likely be extremely difficult to raise through the private sector.

"Banks requiring capital will either need to consider further government support or to sell non-core assets. Financial stocks are likely to underperform in this environment," said Hans Redeker, an analyst at BNP Paribas.

Germany's Deutsche Bank AG was the worst-performing stock on the DAX, down over 4 percent, while in London Barclays PLC dropped over 4 percent and HSBC Holding PLC was down more than 3 percent. In Asia, Mizuho Financial Group slipped 2.0 percent in Tokyo.

On Monday, the Dow Jones industrial average suffered its first drop in three days, falling 51.29, or 0.6 percent, to 8,025.00, while the S&P 500 closed 8.72 points, or 1 percent, to 857.51.

Elsewhere in Asia, South Korea's Kospi retreated 3 percent to 1,300.24. Shanghai's main index was down 0.2 percent, Taiwan's stock measure dropped 1.9 percent while Australia's benchmark was down 0.6 percent

Crucell (CRXL) is up almost 9% to over $20 at this writing, while BioCryst (BCRX) is up 90%. I still think CRXL is worth $35 a share in a buyout, and this news makes it a little more likely that we will see a bid this year.

The trials continue to recruit patients and dose escalation continues beyond the levels at which telomerase inhibition was first detected clinically.

These are significant findings because cancer stem cells are the small sub-population of cells in a tumor that are believed to be responsible for cancer recurrence due to their resistance to standard treatments. We believe we have a drug with potentially broad anti-cancer stem cell activity.”

Therapeutic Targeting of Telomerase for Non-Small Cell Lung Cancer

Lung cancer is the most frequent cause of cancer-related deaths in the U.S. Telomerase is essential for sustained cell proliferation in a wide range of malignant tumors, including 95% of lung cancers. Imetelstat is currently being tested in a Phase I/II clinical trial for non-small cell lung cancer (NSCLC) in combination with standard treatment (carboplatin and paclitaxel chemotherapy).

Data from a companion preclinical study, presented in a mini-talk by Ms. Robin Frink and colleagues from the University of Texas Southwestern Medical Center, showed that continued treatment of NSCLC cell lines with imetelstat inhibited telomerase activity, induced progressive decreases in telomere length, and resulted in cell death. Treatment with imetelstat over several months showed a dramatic decrease in the ability of NSCLC cell lines to form colonies in culture. In addition, there were marked changes in the expression of genes associated with stem cell proliferation. These findings suggest that targeting telomerase therapeutically will require sustained treatment with imetelstat and also provide preliminary evidence that the putative stem cell component of NSCLC is targeted by the drug.

Imetelstat Sodium (GRN163L) Diminishes Self Renewal Capacity and Decreases Tumorigenic Potential of Putative Cancer Stem Cells from Human Breast Cancer and Pancreatic Cancer Cell Lines

Data on the effects of imetelstat on putative cancer stem cells from a human breast cancer and a pancreatic cancer cell line were presented in a poster by Dr. Immanual Joseph and other Geron scientists. The data show that the number of putative cancer stem cells in the human breast cancer cell line MCF7 and their ability to self renew (a key characteristic of stem cells) was dramatically diminished after treatment with imetelstat. In studies of the human pancreatic cancer cell line PANC-1, exposure to imetelstat in culture resulted in the reduction of the putative cancer stem cell population and a significant decrease in the ability of these cells to form tumors in mice.

The Effects of Telomerase Inhibition on Prostate Tumor-Initiating Cells

Prostate cancer is the most common type of cancer in men and has a high morbidity with relatively few chemotherapeutic options for patients with metastatic disease. As in many other cancers, high levels of telomerase activity have also been found in prostate tumors. The data presented at AACR provide a strong rationale for initiating clinical studies of imetelstat in patients with prostate cancer.

Presented in a poster by Dr. Calin Marian and colleagues from the University of Texas Southwestern Medical Center, studies of four human prostate cancer cell lines demonstrated that, as in all tumor types tested to date, the high levels of telomerase activity in these cell lines can be inhibited by imetelstat and that continued treatment led to telomere shortening and a decreased proliferation potential in culture. Once a sufficient shortening of the telomeres was achieved by sustained exposure to imetelstat, the prostate cancer cell lines tested showed an increased sensitivity to the standard chemotherapeutic agents, docetaxel and doxorubicin. Long-term exposure to imetelstat also resulted in a progressive and dramatic depletion of the ability of prostate cancer cell lines to form colonies in vitro, evidence for activity against prostate cancer stem cells.

Putative cancer stem cells, also called tumor-initiating cells, isolated from the prostate cancer cell lines using two different methods (either expression of specific cell surface markers or by dye exclusion properties) were found to have similar levels of telomerase activity to the bulk population and that activity could be inhibited by imetelstat. In addition, the average telomere length of the putative cancer stem cells was similar to the main population and telomerase inhibition led to shortening of telomeres in both populations.

Biologic and Therapeutic Role of Telomere Maintenance in Pediatric Neural Tumor Initiating Cells

Pediatric neural tumors (PNT), comprising childhood CNS tumors and neuroblastoma, are the leading cause of morbidity and mortality among childhood cancers. Telomerase activity can predict survival in PNT and it has been shown that in gliomas and neuroblastomas telomere maintenance governs the unique tendency of PNT to undergo spontaneous growth arrest on the one hand or recur despite maximal resection and chemo-radiation therapies on the other.

Data on the effect of imetelstat on glioma and neuroblastoma-derived cell lines with stem cell-like characteristics were presented in a poster by Dr. Cindy Zhang and colleagues at the Arthur and Sonia Labatt Brain Tumor Research Center and the Research Institute of the Hospital for Sick Children in Toronto in collaboration with Geron scientists. The data show that telomerase is only active in the subpopulation of the neural tumor cells with stem cell characteristics (self-renewal). In such cell lines derived from both glioblastoma and neuroblastoma, treatment with imetelstat resulted in an inhibition of telomerase, a loss of replicative potential, a progressive shortening of telomeres and cell senescence. In addition, cells that were pre-treated with imetelstat were more sensitive to radiation than untreated cells in vitro.

Telomere Shortening in Human Tumor Cells In Vitro and In Vivo Following Treatment With Telomerase Inhibitor Imetelstat (GRN163L)

Presented in a poster by Dr. Ekatarina Bassett and colleagues at Geron with collaborators at the Sidney Kimmel Comprehensive Cancer Center at Johns Hopkins, an alternative and more sensitive PCR-based method (STELA – Single Telomere Length Assay) was used to monitor changes in telomere length in a range of human tumor cells after treatment with imetelstat. Conventional assays (e.g., TRF) tend to underestimate the calculated length of short telomeres and many thousand fold more cells are required to see short telomere populations, whereas STELA enables more precise and sensitive monitoring of short telomere changes.

Using STELA, telomere shortening was observed after treatment with imetelstat in a NSCLC cell line after continuous treatment in culture and in xenograft tumors in treated mice. The onset of telomere shortening was found to be gradual and resulted in an accumulation of shorter telomeres compared to untreated controls. Also using STELA, telomere shortening in myeloma cells was found to be induced by imetelstat treatment in both the bulk and stem cell fractions. Furthermore, telomere shortening in unsorted myeloma cells exposed to imetelstat correlated closely with a reduction in the ability of the cells to form colonies. Finally, STELA was used to detect the distribution in telomere length in primary human tissues (peripheral blood, hair follicles and bone marrow) to demonstrate the potential for this assay to monitor samples from patients treated with imetelstat.

About Telomerase and Imetelstat (GRN163L)

Telomerase is a critical and potentially broadly applicable tumor target. The enzyme is expressed in a wide range of malignant tumors, and its activity is essential for the indefinite replicative capacity of cancer cells that enables their malignant cell growth. Telomerase is absent or expressed only transiently at low levels in most normal adult tissues.

Imetelstat is a short chain oligonucleotide that binds with high affinity and specificity to the catalytic site of telomerase, resulting in competitive inhibition of enzyme activity.
